Mat Tudung Dimulai bercerita tentang seorang anak laki-laki bernama Johari yang bekerja di sebuah hotel. Hotel tempatnya bekerja akan menerima kedatangan juri Forbest yang merupakan lembaga penilai yang akan menilai apakah hotel tersebut layak mendapat tambahan bintang atau tidak. Sayangnya, pihak hotel diberitahu bahwa juri akan datang diam-diam dan melakukan penilaian selama satu minggu. Keputusan ini sungguh membuat marah Abang Gee selaku Manajer hotel. Kemudian Johari bersama rekannya, Sabri mulai curiga bahwa keluarga yang datang menginap di hotel tersebut adalah juri The Forbest. Pasalnya, sifat dan gerak tubuh putri keluarga Dato Wahab dan Datin Mun, Neesa, sangat mencurigakan dan tidak terlihat seperti keluarga sungguhan. Sementara itu, Johari juga mencoba mendekati Neesa namun gagal. Akhirnya Johari harus menggunakan cara menyamar sebagai wanita berkerudung bernama Johana hanya untuk mendekat dan mendekati Neesa.
